It is not necessary to have an actual DNA sample from an historical
person. You only need to have the DNA of enough people who can prove a
direct lineage to be able to identify a common strand. Anybody who
possesses that strand is also a direct descendent. It has been done for
Marie Antoinette and an American company claims to be have done it for
Mohammed.

I don't see anything in what I have written that leads to that conclusion.

You are not the first to claim that. However, while there are strong
oral traditions that allow a number of people to lay claim to the
bloodline of Mohammed, the marriage of Jesus and Mary Magdalene is very
much a fringe theory and there is no definitive track of their
offspring, if any. It is not even possible to identify the bloodlines of
Jesus' siblings.

So why did they need to get a DNA sample from Prinny Philip to prove "Anastasia" was a fake and the bodies buried in the USSR were genuine?

Because they were using the much simpler process of tracing MtDNA from a
living relative, descended from a common mother. HRH The Duke of
Edinburgh is related to the Tzarina through her mother, which allowed
the adult female body and the bodies of the children to be linked to his
bloodline. The Duke of Fife and Princess Xenia Cheremeteff Sfir provided
MtDNA for the identification of the Tzar. Comparison of Y chromosome DNA
and autosomal DNA were also carried out to provide proof that the bodies
were related.

Most people of European ancestry can probably claim descent from
Charlemagne, as well as from the majority of his servants. Once you go
back that far, you are running into seriously huge numbers of ancestors.

Apparently Genghis Khan was quite prolific too:

https://www.theguardian.com/uk/2003/mar/02/science.research

We owe it all to superstud Genghis
Warlord Khan has 16m male relatives alive now, says study

One in every 200 men alive today is a relative of Genghis Khan.
An international team of geneticists has made the astonishing
discovery that more than 16 million men in central Asia have the
same male Y chromosome as the great Mongol leader.

It is a striking finding: a huge chunk of modern humanity can
trace its origins to Khan's vigorous policy of claiming the most
beautiful women captured during his merciless conquest.

'One thirteenth century Persian historian claimed that within a
century of Khan's birth, his enthusiastic mating habits had
created a lineage of more than 20,000 individuals,' said team
leader Dr Chris Tyler-Smith. 'That now appears to account for
around 8% of the men in central Asia.'

Did he have to be a superstud, though? Let's try some overly-simplistic
maths:-

1. Khan lived 800 years ago, which I'll take as 40 generations.

2. Suppose at each generation his offspring produce two new offspring
who survive infant mortality and live long enough to produce two new
offspring ...

3. If all of those offspring avoided interbreeding with each other,
there would currently be around 2^40 currently alive in the latest
generation.

That's a bit more than 100 times the current world population.

Of course, the avoid interbreeding bit is what makes this overly
simplistic. Nevertheless, it explains why the queen might quite possibly
be in some way distantly related to Mohammed. Likewise, Harry, for that
matter.

Looking at it the other way around, this table shows how many ancestors
that sort of progression would result in over 40 generations:

This article considers the question in much more detail and considers
the effects of inbreeding:

The Queen is probably more closely related to me than Mohammed.
